PSMA scFv-CD28-OX40-CD3ζ (3rd) CAR Expression Plasmid Vector Item No.: PCAR-209Shipping: 2 μg Storage: -20°C INTRODUCTION Chimeric Antigen Receptors (CARs) are recombinant receptors that are composed of four regions: an antigen recognition domain (ectodomain), a hinge region, a transmembrane region, and an intracellular T cell signaling domain (endodomain). CAR-T cell therapy uses T cells engineered with CARs to target specific antigen(s) for cancer immunotherapy. Human PSMA (Prostate-Specific Membrane Antigen) / FOLH1 (Folate Hydrolase 1) is a type II transmembrane glycoprotein expressed predominantly in the prostate epithelium, with significantly elevated levels in prostate cancer cells. It functions as a glutamate carboxypeptidase, involved in folate absorption (hence the name folate hydrolase 1) and neuropeptide processing. Due to its high expression on the surface of prostate cancer cells, especially in metastatic and hormone-refractory disease, PSMA serves as an excellent biomarker and target for imaging (e.g., PSMA PET scans) and targeted therapies (e.g., radioligand therapy with ¹⁷⁷Lu-PSMA-617). Outside the prostate, lower expression is found in the small intestine, kidney, and nervous system.
